PCR Using Copy Standards, Example


Figure. 0.5 micrograms per lane of tail DNA and 1X, 5X, and 10X transgene DNA-spiked tail DNA was subjected to PCR with lacZ primers, then electrophoresed on a 4% NuSieve:agarose (3:1) gel and photographed. Three out of six mice were transgenic in this PCR experiment. Inclusion of the copy standards allows us to ensure that our detection is sensitive enough for a single copy integration event so that we do not miss identifying any transgenic mice. We also performed PCR with control primers to ensure that each tail DNA was amplifiable and avoid false negative results.
