iPSC Derivation and Adaptation

Feeder-free defined media (FF-DM) 

iPSCs adapted and expanded in feeder-free media (Stem Flex) – 15 vials (3 colonies, 5 vials each) of undifferentiated colonies delivered to the client. One immunocytochemistry (ICC) image against OCT-4, Nanog, SSEA-4, and TRA-1-81 will be provided.

Fibroblast Derivation from Patient Skin Biopsies

Total of 9 vials = 3 vials of cells collected from each of the attached tissue outgrowths, released from enzyme treatment, cell released from washing after enzyme treatment, and mycoplasma testing.

NPC Derivation

  • iPSC-derived NPC’s from EB and neural rosette generation (10 vials). User supplied iPSCs are validated and mycoplasma free (No ICC and mycoplasma testing required by SCC). 10 vials of NPCs, ICC against  SOX2, PAX6, Nestin, Map2
  • iPSC-derived NPC’s from EB and neural rosette generation (20 vials) User supplied iPSCs are validated and mycoplasma free. (No ICC and mycoplasma testing required by SCC). 20 vials of NPCs, ICC against  SOX2, PAX6, Nestin, Map2
  • iPSC-derived NPC’s from monolayer (10 vials) User supplied iPSCs are validated and mycoplasma free. (No ICC and mycoplasma testing required by SCC). 10 vials of NPCs, ICC against  SOX2, PAX6, Nestin, Map2

NPC Inventory

  • 7545-5B – Monolayer derived from feeder-free 7545-5B iPSC.
  • 7753-5B – Monolayer derived from feeder-free 7753-5B iPSC.

iPSC Inventory

  • 7545-5B – Feeder-free derived iPSC from apparently healthy female fibroblast with normal psychiatric evaluation cultured in defined media (StemFlex or E8).
  • 7753-5B – Feeder-free derived iPSC from apparently health male fibroblast with normal psychiatric evaluation cultured in defined media (StemFlex).
  • 7545-8C – Feeder-free derived iPSC from apparently healthy female fibroblast with normal psychiatric evaluation cultured in defined media (StemFlex or E8).

iPSC Characterization

Feeder-free adapted iPSCs in defined  media (E 8 media) spontaneously differentiated to embryoid bodies (EBs) to test for pluripotency. At day 7-10, EBs immuno-stained against nestin, sox1 (ectoderm); alpha-smooth muscle, brachyury T (mesoderm); and AFP, PDX1 (endoderm).

Lonza 4D Nucleofector

You can schedule an experiment by emailing Veronica Porterfield (vp7p@virginia.edu). Rates per hour, 30 minutes minimum either assisted or unassisted, charged from the beginning of the scheduled time (Rates include use of hoods, centrifuge, incubators and consumables). In addition to the hourly rates, Amaxa P3 Primary Cell 4D-Nucleofecto X Kits can be purchased at cost and availability through the Stem Cell Core or the user can purchase directly from Lonza. For all other Lonza kits, the user must purchase from Lonza.
