RESEARCH SUMMARY:

Over the last 10 years, my attention has focused on the analysis of extracellular vesicles (EVs) as new sources of biomarkers for early detection and investigation of damage to the kidney due to diabetes. I joined Dr Erdbrügger laboratory in 2016 to investigate the role of plasma and urinary extracellular vesicles in hypertension. Not only have EVs become a source of new diagnostic markers, but it is also now agreed that they represent a new cell-to-cell communication system with unlimited potential to develop new therapeutic strategies aimed to achieve tissue regeneration and repair of damage caused by any primary disease. The study of biomarkers relevant in kidney damage due to hypertension and other renal diseases in humans and corresponding animal models of disease has become of paramount importance. An early diagnosis following the first signs of kidney imparment is badly needed to intervene in order to delay the progression of the injury. In parallel, the analysis of urinary extracellular vesicles (uEVs) and plasma extracellular vesicles (pEvs) requires a costant improvement of experimental design and techniques such as tunable resistive pulse sensing (TRSP), nano track analysis (NTA), imaging flow cytometry and other high-sensitivity tools employed to analyze extracellular vesicles. Moreover, the expanding research to understand the physiological role of EVs calls for the need for extreme fine tuning of the biochemical purification tools of choice in order to obtain the highest possible pure preparation of extracellular vesicles by eliminating any confounding carry over of other molecules such as soluble proteins abundantly present in any biofluids. For this reason, designing new purification strategies to minimize loss of precious material from limited amounts of specimen while preserving the integrity of vesicles is another exciting and demanding stream of research currently in progress to test and identify EVs activity pathways.
