Quantitative analysis of confocal FRAP data
With the development of genetic means to labeling proteins in living cells with fluorescent proteins, it has become possible to monitor the kinetics of events such as protein diffusion in the cytoplasm or transient binding events to membranes using biophysical approaches such as confocal FRAP. Importantly, confocal FRAP is widely accessible to an exceptionally large number of experimentalists, and when analyzed using appropriate models, hold the potential to provide quantitative measurements of the on and off rates of binding, as well as directly measure the diffusion rate of the bound and unbound species. Work from my group in collaboration with bio-mathematicians has advanced knowledge in this arena through the development of mathematical models to analyze diffusion and reaction-diffusion events as tools to study intracellular protein and lipid dynamics.
