Macromolecular assembly

Assembly of anthrax proteins into a trans-membrane pore

Subhashchandra Naik, Susan Brock, Narahari Akkaladevi, Jon Tally, Wesley Mcginn-Straub, Na Zhang , Phillip Gao, E. P. Gogol, B. L. Pentelute, R. John Collier, and Mark T. Fisher  dx.doi.org/10.1021/bi400705n | Biochemistry 2013, 52, 6335−6347

Membranes, liposomes, viral like particles

Jakob Wallner, Gabriele Lhota, Dominik Jeschek, Alexander Mader, Karola Vorauer-Uhl Journal of Pharmaceutical and Biomedical Analysis 72 (2013) 150– 154
Used streptavidin probes, bound biotinylated EPO, concentration unclear, saturation within 2 minutes measured binding of liposomes to Epo at 2.5  to 10 mM.

binding of HIV-1 gp120 to liposomes immobilized on aminopropylsilane probes
G.G. Fouda et al Retrovirology 2013, 10:3 doi:10.1186/1742-4690-10-3

using viral like particles as a means of immobilizing membrane proteins. J Dantzler, presentation at fortéBIO using meeting, Seattle, WA 2014; see reference below

Kinetic Analysis of Antibody Binding to an Expressed Membrane Protein on Captured Lipoparticles. fortéBIO Application Note 3. The technique used 150 nm virus like partices, also called lipoparticles from Integral Molecular. This note used biotinylated wheat germ agglutinin to attach the lipoparticles to the probe.

Origin of the Conformational Heterogeneity of Cardiolipin-Bound Cytochrome c Yuning Hong, Julia Muenzner, Sebastian K. Grimm, and Ekaterina V. Pletneva J Am Chem Soc, 134(45), 18713-23, 2012
Attached biotinylated cytochrome c to streptavidin probes. The the authors attached vesicles made from 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) and cardiolipin to the immobilized cytochrome c; vesicle radius was 40 nm or more. Then observed binding of native cytochrome c to the bound vesicles.

Binding of liposomes to synaptotagmin. Marija Vrljic, Pavel Strop, Ryan C. Hill, Kirk C. Hansen, Steven Chu, and Axel T. Brunger Biochemistry, 50 (46), 9998-10012, 2011. Synaptotagmin was biotinylated and immobilised to streptavidin probes with capture levels of 2-2.9 nm. Then liposomes of varying composition and a radius of 50 nm bound to synaptotagmin, which was nearly irreversible. Binding levels of up to 4 nm recorded.

Binding of peptides to proteins

A biotinylated peptide was bound to a streptavidin probe to measure binding affinities of some proteins. Note that current recommendations are to immobilize the protein and leave the peptide in solution. The protein appear bivalent and immobilizing the bivalent component is normally recommended.
Zienkiewicz J, Armitage A and Hawiger J, J Am Heart Assoc, 2(5):e000386, 2013

Biotinylated peptides were bound to streptavidin probes, possibly super streptavidin, and dipped in binding protein in PBS with 0.02% Tween-20, 1 mg/ml BSA.Costin JM, Jenwitheesuk E, Lok S-M, Hunsperger E, Conrads KA, et al. (2010) Structural Optimization and De Novo Design of Dengue Virus Entry Inhibitory Peptides. PLoS Negl Trop Dis 4(6): e721. doi:10.1371/journal.pntd.0000721
Current recommendations are to immobilize the larger binding partner.
